The PhAST Blue equipment has been used to validate, for the first time, the use of viable PCR for the protozoa Acanthamoeba castellani DNA. The dye pre-treatment using a PMA concentration of 200 µM, an incubation time of 30 minute, and photoactivation with PhAST blue for 15 minute, was effective in DNA neutralization of dead protozoa, allowing the detection of both viable trophozoites and cysts of amoebae.
